缺氧诱导RF/6A细胞ILK的表达及其与下游分子表达的相关性
摘要 目的:研究体外缺氧刺激后RF/6A细胞ILK、HIF-1α 、SDF-1及ICAM-1的表达变化,ILK与HIF-1α 、SDF-1及ICAM-1表达的相关性。
方法:体外传代培养RF/6A细胞,氯化钴(CoCl2)150μmol•L–1对RF/6A细胞进行体外缺氧干预,干预时间为0 h、6 h、12 h、24 h、48、72 h,RT-PCR法、Western blot法检测ILK、HIF-1α 、SDF-1及ICAM-1的表达变化,用LY294000抑制ILK的活性后再次检测缺氧后ILK、HIF-1α 、SDF-1及ICAM-1的表达变化。
结果:缺氧刺激后,ILK与HIF-1α 、SDF-1、ICAM-1在RF/6A细胞上均有表达,缺氧早期呈上升趋势,干预后12~24h表达最强,后呈下降趋势;但这种上升趋势都因为ILK的活性受到抑制而被抑制。
结论:缺氧可促进RF/6A细胞表达ILK、HIF-1α 、SDF-1及ICAM-1, ILK与HIF-1α 、SDF-1及ICAM-1的表达呈正相关性。

[外文]Expression and correlation of ILK and downstream molecules in
RF/6A cells under the hypoxic condition
ABSTRACT Objective: To investigate the expression of ILK , HIF-1α ,SDF-1 and ICAM-1 in RF/6A cells under the hypoxic condition and the relationship between ILK and HIF-1α ,SDF-1 and ICAM-1.
Methods: Cultured RF/6A cells were maintained with Cocl2 for 0h、6h、12h、24h、48h、72h respectively in vitro. The expression of ILK, HIF-1α,SDF-1 and ICAM-1 were quantified by Real Time PCR and Western blot. To quantify the expression of ILK, HIF-1α,SDF-1 and ICAM-1 in RF/6A cells under the hypoxic condition after the activity of ILK was inhibited with LY294000.
Results:Under hypoxia condition,ILK , HIF-1α ,SDF-1 and ICAM-1 co-expressed in RF/6A cells. The expression levels of the four factors were all ascended and reached the summit after 12~24h by hypoxia while descended after 72h,However, this upward trend was inhibited after the activity of ILK was inhibited .
Conclusion:Hypoxia can promote the expression of ILK , HIF -1α , SDF -1 and ICAM -1 in RF/6A cells. The expression of ILK and HIF - 1α , SDF -1 and ICAM -1 was positively correlated .
